Incorrect. It's not Google Play credit, it's Google Pay. Different beasts with similar names (so Google!)
Google Pay should be accepted at any store that accepts payment by Paywave style Credit Card assuming you have an appropriate device (phone or smartwatch) that is set up for Google Pay.
Once you've applied the credit to your Google Pay account, you'll have a total of $110 available to spend.
It's not Google Play credit, it's Google Pay

If you look at the gift card do they have a pin you need to enter? if yes it's 2 transactions
Not 100% sure, but I suspect they are two separate card numbers, right? So you would have to add them to your google wallet separately. Then when paying, you have to select a card, so I can't see how you'd be able to do it in one transaction?
Any instore purchase should allow you to do multiple payment methods, eg I recently bought an $800 item with 4 x $100 egiftcards and the rest my credit card at JB Hi-Fi. Online, jb hi-fi allow up to 5 gift cards apparently. It would depend on the merchant I suppose.
