So I've been curious about getting into the Ereader scene but have a few questions before I take the plunge
-Are the ebook stores interchangeable? Ie. If I purchase one from Amazon's Kindle store, would it be compatible on a Nook and vice versa?
-How does multiple device use work with the same account? Can you have the same books duplicated onto more than one at the same time?
And lastly,
-What is the best Ereader on the market now? I was eyeing a Kindle 3 a while back and with the exchange rate, it'll be <$140 AUD, but the new Nook is out too at the identical pricepoint. Again, if all the purchased ebooks are compatible, would it be possible to read on X device if they were purchased on the competitor's store? Is there any new Kindle model on the horizon I should be worried about?

Generally no. But there is software out there to help convert ebooks to different formats for different readers. Have a look at an e-library management software called Calibre. It's free and very well regarded (I use it myself).

I had the same debate a couple months ago, ended up going with the 1st ed Nook. Mainly for the wider ebook formats it supports (Kindle pretty restrictive). IMO don't bother with the Nook colour as you're better off just getting a tablet.
New Nook would be even better, esp the battery life (1st ed Nook only lasts about a week or two). Catch is B&N won't ship internationally so you'll need friend/relo/forwarder to help out.
